Software:Article Title: Clinical isolates of Staphylococcus aureus exhibit diversity in fnb genes and adhesion to human fibronectin. Article Snippet: Staphylococcus aureus is a leading cause of communityacquired and nosocomial sepsis.1–3 The effective treatment of such infections is threatened by the continuing rise in rates of nosocomial methicillin-resistant S. aureus (MRSA) sepsis,4,5 compounded by the recent emergence of MRSA with intermediate sensitivity to vancomycin.6–9 In consequence, there is a growing need for alternative approaches to prevention and therapy.. Since bacterial adherence plays a key role in the pathogenesis of S. aureus infection,10–12 the pursuit of an in-depth understanding of this process is one element in the development of such strategies.. The fibronectin-binding proteins (FnBPs) of S. aureus have been implicated in the pathogenesis of prosthetic material-related infection by studies that have shown them to promote bacterial attachment to fibronectincoated plastic in vitro,13 human intravenous catheters ex vivo,14,15 and coverslips explanted from the subcutaneous tissue of guinea pigs.13 Staphylococcus aureus 8325-4 has two fibronectin-binding proteins, FnBPA and FnBPB, encoded by the closely linked genes fnbA and fnbB,16,17 both of which contribute to adherence to fibronectincoated surfaces.13 These proteins are described in terms of regions.16,17 When FnBPA and FnBPB are compared, the regions A share 45% amino acid identity, while regions C, D, W and M are 95% identical.10 The ligand-binding activity is located in region D, which is composed of three consecutive repeats of 37 or 38 residues (designated D1, D2 and D3), plus one incomplete repeat (D4).16 The fibronectin-binding proteins of clinical isolates have not been characterized extensively. |
